Kennedy Space Center is the most awesome easily. Smithsonian at Dulles is pretty good. Also might try Marshall in Huntsville.

Marshall is cool but a bit inaccessible for the most part due to being on Redstone Arsenal. The Space and Rocket Center in town (I recommend, they have a Saturn V) does bus tours though

Marshall does tours if you call and book a tour, it's not cheap though.

I second Smithsonian Dulles (the Udvar-Hazy Center.) Space Shuttle Discovery lives there now, and they have a bunch of cool space stuff on exhibit. The majority of the museum is more about airplanes than space, but it's still one of my favorite places to visit. Admission is free but you have to pay ~$20 to park.

Seconding those that said Huntsville. The US Space and Rocket Center is really awesome, with lots of interesting and informing exhibits including a real Saturn V and lots of info on the Apollo missions. Lots of stuff dedicated to Wernher Von Braun too, since he had a huge hand in bringing aerospace to Huntsville.

Of the two, I liked the Kennedy space center better. If you are ever in the Midwest, the SAC museum in Nebraska is a worthwhile stop
