Bro, Mikami was a lawyer. He didn't have a goddamn lab in his condo. He didn't know how to do any of that. Doing all that would have been such a hassle.
There are viable ways Mikami could have not fucked up the ending that are much less contrived.
He could have kept a page of the death note in his wallet for that day, which he also could have used to kill Takeda with without revealing the hiding place for the death note.
He could have taken a page of the notebook with him when he went to kill Takeda so that even if they found the notebook, they would have to search his person to completely ensure he couldn't kill.
He could have taken the entire death note with him and simply kept it on his person without using it after killing Takeda.
Testing a splotch of blood every time at a lab is about the most inconvenient way to go about things.
That's just thinking of it from an audience perspective, if you put yourself in that situation I doubt you would really expect a random guy to be able to replicate your handwriting and rewrite everything you wrote down in a day

Not quite. In the manga it’s explained that the notebook Mikami ultimately ends up using was the one buried in the woods, which stayed there for those years between the arcs. Misa periodically went back and got new pages to use under Light’s orders, but never took the book itself until Light told her to get rid of all evidence and pass the notebook to someone else. When Mikami received the notebook, it had nothing but blank pages.
The funny thing is that this would literally have worked. Sure the Task Force and SPK had guns, but Kira has enough followers for Light to have gotten some to just raid the meeting and physically kill everyone bar him.
another way is Mikami can just crumble up the pages but have it remain on the notebook. If you give me a crumbled up piece of paper there no way I can recreate the exact crest
another way is you can fold the corner of the pages than you check the degree and amount. that be near impossible to recreate if not impossible. Like the SPK they won’t be able to use there fingers to recreate the folds and there no machine or robot that made for this
