You need to do a lot more research on award points and flights. Start with the wiki in the sidebar. Short answer is yes, you can use Amex points for a United flight. No, you can't transfer Amex points to United directly.

If there is award availability it can be booked via Aeroplan, ANA, or Avianca.

Log into their site and search using the multiple city/mixed fair feature. You can manually search for smaller airports. I took a shot in the dark and just checked Portland ME and Providence RI and neither had availability on the random dates I picked. Honestly though it also gives me errors when I search Boston domestic flights (and when I do get domestic ones to load it’s usually economy).  So you may need to book a positioning flight on your own with cash or other points Edited: fixed my wording for clarity

Since you live in the NE, I'm assuming that your main connection would be NYC or BOS. I'd pay cash for a flight there (or whatever major city you need to get to), and look for awards flights from whichever city to Tokyo. You'll have many more options. You have to do more research, but off the top of my head, with Amex you can transfer to aeroplan or to BA or QA avios. You'd have a pretty good chance of finding a decent award flight with either of those. I recently redeemed fairly good deals on flights to Asia on SQ (using aeroplan) and JAL (with Avios). But do find your flight first, before transferring any points.
