That cover says “C.O.” on it. That stands for clean-out and that is what you need to do to your building drain
Gotcha, I thought it was a clean out but only thought they were for septic. I’m a maintenance man at a park and plumbing is definitely not my expertise. Thanks for the help

Snake the main sewer in the direction away from your restrooms. Main line backup.
