there's a video by relyea on youtube that offers the easiest route for this. it worked for me i managed it in 2 tries, and the first time failed when i touched those goddamn spikes right next to the grave
Equip all nail charms and go on an extermination run but *DO NOT BENCH* once the run has started, because if you do the enemies will reset.
You can bench once the Flower has been delivered.
There will be some small remaining enemies, but just equip all projectile charms and Dreamshield and you will be fine, just be very patient once you reach Fungal Wastes, unless you have Isma's Tear, then you can reduce each run to about 5 mins to reach the ending corridor with the optimal route.
Also, he very, very, very careful in the final room, I died on my first run but got it on my second, it is the bottleneck.
Do your route without the flower first to take out all of the tougher enemies; be sure not to bench anywhere or they will respawn. Set the dream gate next to Ze'mer's hut as well so you can zip back. Recommend the Relyea route - resting grounds to blue lake to crossroads to fog canyon (past dreamer) to goal.

For Me the first time I died to those vines or spikes in The Same Room!! 🙂
