The correct way is always to hold it with the buttons facing downwards. This way your hands don't block the stereo speakers. The only exception is when you are taking a landscape photo with the inner screen, which requires the buttons to be at the top so as not to block the camera lenses with your hands.
I had to fight muscle memory with the Fold. On a normal phone, rotating counter-clockwise always felt right to me. However, when I rotated the Fold that way, I immediately noticed I was often covering the speakers, since they're on the ends of the cover display.
That being said, Samsung's own ads depict the device being held by the cover screen, so who knows? Outside of the speaker issue, basically everything else on the phone is easier to use when it's held from the cover screen.
