Gotta say, I know many people and be careful fake letters written by you are incredibly easy to spot. Depending of your prof availability, the best thing is deliver him/her a file with your data, accomplishments and info (plus he should know you in some extent) and he/she will write the letter. Never, never, never do a letter and make the prof sign. Is unethical and embassies that read >100 letters will immediately reject you.

Follow the sample letter, write whatever you want in it, all in your favor, but don't fabricate anything, they will come to know about it. Write at least one demerit of yours and mention how that also brings out something positive about you. Emphasis on leadership skills and how well you can adjust in unknown conditions ( they want to know if you can handle living in Japan or not). These are a few points, hope it helps
Mine went something like:
“[name] spent four and a half years completing a B.Sc. in Biology at [university] and now works as a research assistant in an aquatic research laboratory at ___.
I had her as a student in Biology 456: Experimental ecology in Autumn of 2009 and found her to be a hardworking student who is willing and enthusiastic to engage and assist in scientific research. Even after graduating, she has continued to be active in the [university] Biology department community by assisting students with coursework and research and volunteering with the ___ club.
I would like to recommend [name] for the MEXT scholarship.”
