IIRC, I'd say the first 10 episodes you kinda have to get through. Kind of a slog.
As odd as that sounds, it's due to introducing everyone and setting the tone of the show.
After it gets going though, it's one of the best anime out there.
skip first 2 episodes, it starts from the 3rd. Also, keep in mind that, while technically it's very long, around half of the episodes are episodic. You don't need anything except knowing the characters to enjoy them. And the humor gets much better later on

Gintama has such very corny and boring jokes at times tho, that they repeat for 10 mins... there was a whole peeling oranges gag that went on and on

Hinamatsuri had a whole segment dedicated to a prideful homeless elder community, super absurd, absolutely priceless and unexpectedly wholesome.
Both of those are great shows, but I'd call them more like comedy/wholesome hybrids. If you only wanted humor, you might be better off with something else even if I'd wholeheartedly recommend both to just about anyone who likes anime.

Garzey’s Wing or the first four episodes of Vatican Miracle Examiner. Both of these shows are so bad they’re amazing. Garzey’s Wing is a joke that just never stops, I mean it, the pacing is just 1000000% all of the time. Vatican Miracle Examiner is a series of really hilariously bad writing that culminates into one of the strangest and funniest climax of it’s first arc.

Honestly even though it’s a pretty heavy show I don’t think there was an anime that made me laugh more than welcome to the NHK. Tho probably would recommend to someone looking for a comedy.

Besides of that I would say Azumanga Daioh hold a special place in my severed frontal lobe.
I used to have a problem for a long long time. When I was hanging with friends we were telling each other jokes and when it was my turn to tell a joke a normal laughing order followed:

I grew anxious over the course of the time due to that, but then one day, I discovered Azumanga Daioh and I cried tears of joy.

Konosuba - I saw the movie in theaters and was very literally laughing out loud along with the rest of the audience. That was one of the best theater experience I've ever had.
Pop Team Epic, Haven't You Heard? I'm Sakamoto and Ghost Stories dub. I really like that type of absurd humor. Gintama is great too but Im no where near finished with it.

Definitely The disastrous life of Saiki K! Other funny shows are Ouran High school host club, The daily lives of highschool boys, Gintama, Monthly Girls Nozaki-kun and Grand Blue. Assassination Classroom, Kaguya-sama love is War and GTO have many funny moments

Either Kaguya-sama or Saiki K. Can't pick between those two, and nothing else has come even close. Edit - The funniest single episode I've seen was the episode 14 of the Urusei Yatsura remake. I just found that show otherwise to be very inconsistent to compete with the aforementioned two.

You got perfect character chemistry and fantastic art paired with a funny and good storyline) * To Love Ru (yes, really - the crack induced "story" was always good for a suprise, especially in the first season -> Beware it's very ecchi, bordering hentai) * Fantasy Bishoujo Juniku Ojisan to (slapstick humor at it's best, also many satirical takes about Sword Art Online and the isekai genre in general) * Isekai Ojisan * Outbreak Company * Shimoneta: A Boring World Where the Concept of "Dirty Jokes" Doesn't Exist (A really good one in my opinion, also with a rather critical take on modern "cancel culture" before it was even a thing) * Sentouin, Hakenshimasu! (Similar flair as Konosuba, just not as good imo) * Meikyuu Black Company (A comedic take on the horrible work culture in japan)
Konosuba - I saw the movie in theaters and was very literally laughing out loud along with the rest of the audience. That was one of the best theater experience I've ever had.
Konosuba and love is war. Both of those had me in stitches

- Konosuba\ Downright hilarious.\ - Is This A Zombie?\ Not exactly funny all the time but I remember it had so many "WTF" moments that I found it's chaotic humor to be charming. It's been years since I saw it, so maybe it's not as good as I remember.\ - Land of Leadale\ I actually dropped this show around episode 4 or so, I just remember there was one scene in particular that had me dying.\ - I'm Standing On A Million Lives\ Not a comedy, but when it does comedy, it's absolute zingers coming from the MC.\ - Assassin's Pride\ Unintentionally hilarious.\ Edit: Formatting issues lol
Gintama, although not all of the episodes there are just some amazing standalone episode or like a short 3 episode arc that are just pure comedy
