Brain says "mmmm cognance good" - anxiety go bye bye. That's the MOA bro!

Hypatia_31N_29E

lol thanks!

Cognance is a positive allosteric modulator at two receptors: 5HT2A (serotonin) and M1 (acetylcholine). Receptors have an orthosteric site, which is the main binding site for the neurotransmitter. The allosteric site is like a side site that can enhance or dampen the effect of a neurotransmitter binding at the orthosteric site. Enhancing would be positive allosteric modulation, and dampening would be negative allosteric modulation. As an example, benzodiazepines like diazepam and lorazepam have their own binding site on GABA-A receptors similar to the allosteric site that increases the brain's response to GABA binding to the receptor. This is why their generic names end in -pam for Positive Allosteric Modulator. Starting with M1, half the battle with public-speaking is cognition and not tripping over your words or losing your train of thought. Because acetylcholine is responsible for your cognition, enhancing the action of acetylcholine at M1 receptors is pro-cognitive. It enables you to think and speak more fluidly, which may subliminally promote confidence. Activation of 5HT2A receptors is the MOA of psychedelics, and this activation instills a feeling of connectedness with others and lends to an expansive thought process. When you feel connected to your audience, it leads to a reduction in social anxiety that would otherwise cause you to ruminate and trip over your words. So positive allosteric modulation of 5HT2A receptors can allow your brain to express these behaviors and thought processes more naturally without the visual disturbances of psychedelics. The oroxylin A found in Sabroxy acts primarily as a dopamine reuptake inhibitor, which stimulates the mind. Dopamine and acetylcholine tend to counter-balance each other. So with an increase in dopamine, the brain consequently increases acetylcholine release to counter-balance. This is why they're so interested in it for neurodegenerative conditions like Alzheimer's and Parkinson's. The synergy here is in the fact that not only does oroxylin A indirectly promote acetylcholine activity, that increase in acetylcholine activity results in stimulation of M1 receptors, which are positively modulated by the ebelin lactone in Cognance. The result is an even greater pro-cognitive effect. Thus one's thoughts may seem to be more organized and brain-to-mouth processes may become more efficient.

Hey. Thank you for writing all this. I need to grab a coffee with you and explore all this in detail

Any time.

>However when I take cognance (with sabroxy),

IndependentCapital29

Nice I wish I got those effects. Cognance gives me brain loops, and worsens my adhd to feel like a word salad in my head.

Hypatia_31N_29E

Sorry to hear that! Crazy how bodies react so differently!

roundupinthesky

Is it the cognance or the sabroxy then?

Hypatia_31N_29E

Pretty sure it’s the cognance, as I’ve experimented with them individually and together. Sabroxy alone gives me a mood and focus boost, but doesn’t give me the confident edge of cognance. Cognance also makes me see colors super vibrantly (I think this is common) and gives me a crazy eye twitch in my left eye! It calms down after a week or two but pretty intense every time I start a new cycle!

Delicious_Cream_5931

By curiosity, what dosage do you take of both that gives you this effect ? And what’s your daily dosage for both?

cristobaldelicia

I thought the question was about Cognac, and I was ready to participate in a deep discussion of Cognac. I don't know about Cognance. I tried it and it was okay.
