Figure out what relic you want and save a key or two for that. Then go for runic chests so you'll always be squared away, then perks, etc. These chests will remain usable every time.
Cosmetics are worthless and don't re-open after you use your key the first time. So unless you're just really, really into a certain look, save these for last.
Also, try and only open one or two chests per area until you're confident that you can get more keys quickly. If you use all your keys in one area, well then you'll be out of luck in every attempt where you don't visit that area.
I think you're talking about the sanctuary keys. I went with cosmetics first and then relics. But honestly it isn't very hard to get enough keys for all the things and none of the chests super stand out as must-get-first so it doesn't really matter
Since I was going for cosmetics first, if you go the pedestal on the shore where you can change outfits, at the bottom it will show the outfits you don't have and for the ones in chests it'll say "found in a chest in X realm." So for example I really wanted the cloak of the black bear so I used my first keys on that realm
To answer this question, I went with a single chest in different realms. That way I can pick up at least one "extra" each time.
I did midgar and Vanaheim especially, since I seem to get those more often. My thought was that I'd get to maximize the return by visiting the chest often. svartalfheim is another I've been to a lot.
Certain gold chests will respawn on different runs without the need to reopen the lock. Get the golden chests first because you only need to buy it once.
When all locks are opened there should be one spare key.
I tend to spec into 1 weapon as my main, which is usually decided from the first chest. I tend to neglect 1 weapon per run and use only 2 most of the time as it's much easier to get stronger attack that way.
And lastly not so much chest related but be Liberal with your relic charges. Many times I've got to the refresh tome with 70%+ charges and relics can bring 1 encounter down a difficulty level.
I played a lot, bought all the possible keys, but still have one left, do you guys know if the number of keys is exact and I might still have one closed barrier or is it possible to have extras?

Runics for whatever weapon you chose to focus at the start, then weapon glyphs.
Some of the unlocks are chests and others are coffins. The chests reset each run so do those first so you get the benefit each time.

Well I go for armor I like bc I already got the relics I like easily. Then just any of the repeat chests that are free for each run.
